Daraxonrasib, also known as RMC-6236, is a targeted cancer medicine that inhibits RAS signalling. RAS proteins can drive cancer cell growth and survival when abnormally activated. By inhibiting RAS activity, daraxonrasib helps interfere with pathways involved in tumour growth.

Daraxonrasib is an oral targeted anticancer medicine. The US FDA approved it for certain adults with metastatic pancreatic adenocarcinoma. The usual recommended dose is 300 mg once daily, so a 100 mg tablet is a strength of the medicine, not necessarily the patient's total daily dose.

It is available as an oral tablet. Treatment should be initiated and monitored by an oncologist experienced in cancer-directed therapy.

  • Daraxonrasib is approved in the United States for adults with metastatic pancreatic adenocarcinoma who have received at least one previous systemic treatment or who are not candidates for multi-agent systemic therapy.

  • Daraxonrasib is also being investigated in clinical trials for other RAS-related cancers, including certain lung cancers and other solid tumours. These investigational uses should not be confused with the approved indication.

  • Strong CYP3A inhibitors: Some combinations must be avoided; others require a dose reduction.
  • Moderate CYP3A inhibitors: May require a dose adjustment.
  • P-gp inhibitors: May increase daraxonrasib exposure and require dose adjustment.
  • CYP3A inducers: May reduce drug exposure; strong inducers should generally be avoided.
  • Cyclosporine A: Avoid concomitant systemic use.
  • P-gp substrate medicines: May need to be administered at least four hours apart.

Tell the oncologist about all prescription medicines, over-the-counter products, vitamins and herbal supplements. Never adjust Daraxonrasib's dose yourself to manage an interaction.
