Talazoparib capsules contain the active ingredient Talazoparib, a targeted anti-cancer medication belonging to the class of PARP (Poly ADP-Ribose Polymerase) inhibitors. It is used in the treatment of certain types of advanced breast and prostate cancers associated with BRCA or other homologous recombination repair (HRR) gene mutations.

Talazoparib works by preventing cancer cells from repairing damaged DNA, leading to cancer cell death while limiting damage to normal cells.

Available strengths:

Talazoparib 0.25 mg capsules

Talazoparib 1 mg capsules

These lower strengths are generally used for dose adjustments or in patients requiring modified dosing due to side effects or kidney function considerations.

Talazoparib is prescribed for:

Germline BRCA-mutated (gBRCAm), HER2-negative, locally advanced or metastatic breast cancer.

HRR gene-mutated metastatic castration-resistant prostate cancer (mCRPC) (as per physician recommendation and regional approval).

Treatment should only be initiated after genetic confirmation of mutation status.

Talazoparib inhibits the PARP enzyme involved in DNA repair. Cancer cells with BRCA mutations already have impaired DNA repair mechanisms. By blocking PARP, Talazoparib further prevents repair, causing cancer cells to die.

Dosage & Administration

The usual recommended starting dose is 1 mg once daily (as prescribed by the oncologist).

0.25 mg and 0.1 mg capsules are commonly used for dose reductions in case of side effects.

Swallow the capsule whole with water.

Can be taken with or without food.

Do not crush, chew, or open the capsule.

Possible Side Effects

Common side effects include:

Anaemia (low red blood cells)

Neutropenia (low white blood cells)

Thrombocytopenia (low platelets)

Severe bone marrow suppression

Myelodysplastic syndrome (MDS)

Contact your doctor immediately if you experience unusual bruising, bleeding, persistent fever, or severe weakness.

P-glycoprotein (P-gp) inhibitors.

Other chemotherapy agents.
